cI434 repressor from phage 434 optimized for E. coli cI (+LVA)
Optimized cI434 repressor with LVA tag from phage 434, with preferential codon usage for improved expression in E. coli.
Usage and Biology
Repressor protein cI from bacteriophage 434. cI434 has a higher affinity for OR1 and OR2 operators, where binding positively regulates the pRM434 promoter. On the other hand, when bound to OR3 cI434 represses the promoter. Repressor binds to each operator site as a dimer; however, due to a low binding affinity, a higher concentration of the protein is required. Thus, c434I can act as both an activator -at low concentrations- and a repressor -at high concentrations- on pRM434.
